It's still 140 characters, but the new Twitter is also a lot more. Apart from the Twitter bird (I miss the old one) the "new, re-engineered Twitter.com" has added a bunch of new stuff to the user interface.
New design
Twitter has made the timelines cleaner, and added a rich details pane . They've also added 'infinite scroll' - no more clicking 'more'.

See photos and videos directly on Twitter (instead of going to another site); Twitter's tied up with DailyBooth, DeviantART, Etsy, Flickr, Justin.TV, Kickstarter, Kiva, Photozou, Plixi, Twitgoo, TwitPic, TwitVid, USTREAM, Vimeo, yfrog, and YouTube for this.

Since every tweet has a story behind it, related content includes information related to the author or subject. Replies to the tweet, other tweets, some geo-taggery other integration thrown in.
